Whenever you play a Skill, gains 2 Strength.

The Test Subject boss in Glory starts with 2 stacks (3 at Ascension 9+). Whenever a Skill card is played, it waits half a second and then gains Strength equal to the stacks. The code checks only the card's type, never who played it, so in multiplayer every player's Skills feed it. The Strength is permanent.

Notes

Block cards are Skills, so every Defend hands it 2 Strength. Attacks and Powers are safe. It is never removed across the boss's respawns; only Adaptable and Painful Stabs come off. Build your defense from Powers or Attacks that block, or simply race it.
