At the start of the player's turn, Turret Operator gains 25 Block.
Rampart belongs to Living Shield, which puts 25 stacks on itself as the fight begins. At the start of each regular player turn it grants that much Block to every Turret Operator in the fight, never to itself. The Block is flat, ignoring any modifiers on the shield. Killing the Living Shield ends the power, and with it the operator's free wall.

When the Block lands
The Block lands at the start of your turn, so the operator opens every one of your turns with 25 fresh Block on top of whatever it kept.
Rampart does not fire during an extra turn granted to a player, only on the normal turn start.
In multiplayer the amount is multiplied by the player count.
The practical answer is to kill the Living Shield first or lean on damage that ignores Block.

In co-op an enemy receives more of it: the amount rises with the number of players.
