Kill the Act 3 boss as The Necrobinder and Spireborn is unlocked at once, mid-run, as soon as the boss room is won.
It is the third boss of a Standard run that matters, not the ending screen; Daily and Custom runs do not count. The slot is revealed by Life and Death, and Spireborn reveals nothing further.
