You gain 12 Block, and the amount is fixed: Dexterity does not raise it. Like any Block it vanishes at the start of your next turn unless something such as Barricade keeps it. In a co-op run you can throw it to a partner instead of drinking it yourself.

When to take Block Potion

A cheap panic button, so there is no reason to hoard it. Drink it when a single hit would take you low and your hand cannot cover it.
