What it unlocks
The Timeline lists no card, relic or potion for it, but one event waits on it: Colorful Philosophers is removed from the event pool of every act until The Spire is opened on the Timeline. Nothing else changes: no cards, relics, potions or modes. Chapter 9 of 9 in Magnum Opus, the story's ending, in the Blight era.
How to unlock The Spire
The Spire unlocks from the score bar as the last of the 18 score epochs, at 2500 points.
Score from each Standard run accumulates in one bar across runs; this is the final bar, so once it fills the bar disappears and later score does nothing. Its slot is revealed by Arise, the sixteenth score epoch.
The Spire reveals no further slots.
